Mechanism of action

IGF-1 LR3 is an 83-amino-acid analogue of human IGF-1 with an N-terminal 13-amino-acid extension and a Glu→Arg substitution at position 3. These modifications dramatically reduce binding to IGF binding proteins (IGFBPs), extending plasma half-life from ~15 minutes to 20–30 hours. IGF-1 signals through IGF-1R and secondarily IR to activate PI3K/AKT and MAPK/ERK pathways, driving protein synthesis, glucose uptake, cell proliferation, and anti-apoptotic signaling.
